Citation: Stamp D. (2016). Proposed Use of Deoxycholic Acid (DCA) and Ursodeoxycholic Acid (UDCA) in a Treatment Regimen for Barrett’s Esophagus. M J Canc. 1(2): 007. INTRODUCTION Huo et al [1] first placed 250 uM of the hydrophobic bile acid (DCA) into the esophagus of patients with Barrett’s esophagus (BE) and left it there for 5 min. The DCA was dissolved in alcohol. Subsequent examination of the biopsies taken from the esophagus showed that the DNA was damaged and the nuclear factor NF-kB was activated. Additionally, many other pre-cancerous lesions were activated. Similar treatment with the hydrophilic bile acid (UDCA) was ineffective, suggesting that UDCA could be an anti-carcinogen.

In Barrett's esophagus patients and Barrett's cell lines, ursodeoxycholic acid increases antioxidant expression and prevents DNA damage by bile acids.

Hydrophobic bile acids like deoxycholic acid (DCA), which cause oxidative DNA damage and activate NF-κB in Barrett's metaplasia, might contribute to carcinogenesis in Barrett's esophagus. We have explored mechanisms whereby ursodeoxycholic acid (UDCA, a hydrophilic bile acid) protects against DCA-induced injury in vivo in patients and in vitro using nonneoplastic, telomerase-immortalized Barret...

Ursodeoxycholic acid can suppress deoxycholic acid-induced apoptosis by stimulating Akt/PKB-dependent survival signaling.

The nontoxic bile acid ursodeoxycholic acid (UDCA) is reported to be an anti-apoptotic agent with efficacy against a variety of death stimuli including the cytotoxic bile acid deoxycholic acid (DCA). To gain insight into this anti-apoptotic property, we tested UDCA for its ability to protect the colon carcinoma-derived cell line HCT116 against DCA-induced apoptosis. We found that UDCA could sup...

Deoxycholic acid causes DNA damage while inducing apoptotic resistance through NF-κB activation in benign Barrett's epithelial cells.

Gastroesophageal reflux is associated with adenocarcinoma in Barrett's esophagus, but the incidence of this tumor is rising, despite widespread use of acid-suppressing medications. This suggests that refluxed material other than acid might contribute to carcinogenesis. We looked for potentially carcinogenetic effects of two bile acids, deoxycholic acid (DCA) and ursodeoxycholic acid (UDCA), on ...

Novel channel structure of bile acid-guest inclusion complex formed between ursodeoxycholic acid and phenanthrene.

The crystal structure of inclusion complex between ursodeoxycholic acid (UDCA), and phenanthrene has been determined. UDCA molecules formed hydrogen bond network to provide the channel structure along b axis, and phenanthrene molecules were accommodated in the cavity with a stoichiometry of 1 : 1 molar ratio. The channel structure observed in the UDCA-phenanthrene complex was significantly diff...

Ursodeoxycholic acid inhibits translocation of protein kinase C in human colonic cancer cell lines.

Deoxycholic acid (DCA) has been implicated in colonic carcinogenesis through effects mediated by protein kinase C (PKC) activation. By contrast, ursodeoxycholic acid (UDCA) is reported to reduce colon cancer incidence in ulcerative colitis. The aim of this study was to investigate whether UDCA modulated DCA-induced PKC isoenzyme translocation to its site of activity. HCT116 cells were treated w...
